Adenosine-triphosphatase activity on the plasmalemma and tonoplast of isolated mesophyll protoplasts, isolated vacuoles and tonoplast-derived microsomes of the Crassulacean-acid-metabolism plant Kalanchoe daigremontiana Hamet et Perr., was localized by a cytochemical procedure using lead citrate. Enzyme activity was detected on the cytoplasmic surfaces of the plasmalemma and tonoplast. The identity of the enzymes was confirmed by various treatments differentiating the enzymes by their sensitivity to inhibitors of plasmalemma and tonoplast H+-ATPase. Isolated vacuoles and microsomes prepared from isolated vacuoles clearly exhibited single-sided deposition on membrane surfaces.Abbveviations CAM Crassulacean acid metabolism - H+-ATPase proton-translocating ATPase  相似文献

叶绿体中存在着与细胞分裂素(CTK)专一结合的蛋白质。这一蛋白与6-苄氨基嘌呤(6 BA)的亲和力很强,解离常数达3.7×10~(-8)mol/L。最大结合量为10.7 pmol 6 BA/mg蛋白,Seatchard分析表明只有一类结合位点。不同的叶绿体纯化步骤对CTK结合蛋白的活性有不同的影响,分离步骤少而快速的差速离心法可以得到具有较高结合活性的叶绿体。叶绿体经分离纯化后,低温保存时的结合活性较稳定,-20℃以下可以较长期保存。用EDTA预处理叶绿体,不降低CTK结合蛋白对6 BA的结合活性,而用高浓度的NaCl处理,可以使叶绿体结合6BA的能力明显下降。这说明EDTA不能使CTK结合蛋白从叶绿体膜系统表面解离,而高浓度NaCl则有这种可能性。  相似文献

This research was conducted on a captive group of eight wild boars: Four wildborn adults (one male and three females) and four piglets (one male and three females), born in captivity two months before the beginning of the study. They were housed in the Rome Zoo in a 1,000 sq m enclosure furnished with trees, mud pools, a water basin, and two small barns. Data on eight behavioral states (sleeping, resting, rooting, eating, walking, standing, scratching, and nursing) were collected by instantaneous sampling at 10-minute intervals. The spatial location of each subject was also noted, as well as closeness to conspecifics. Results showed that adults and piglets differed in their activity budgets. Activity patterns showed a significant correlation with the mean temperatures. Furthermore, there was a significant difference in the use of the exhibit areas between adults and piglets. This study suggests a social structure in which the core is constituted by females and their yearlings. The nonbreeding female has a more marginal position than the breeding ones. Finally, the male tends to be even more peripheral and solitary. Our results are partly consistent with the data obtained in studies on free-ranging wild boars (Mauget, 1980; Dardaillon, 1984).  相似文献

The mechanism of therapeutic activity of recombinant murine interferon-gamma (rMu IFN-gamma) and the IFN inducer polyinosinic-polycytidylic acid solubilized with poly-L-lysine in carboxy methyl cellulose (pICLC) in treating metastatic disease was investigated by comparing effector cell augmentation with therapeutic activity in mice bearing experimental lung metastases (B16-BL6 melanoma). Effector cell functions in spleen, peripheral blood, and lung (the organ with tumor) were tested after 1 and 3 weeks of rMu IFN-gamma or pICLC administration (intravenous, three times a week). In these studies, natural killer (NK), lymphokine-activated killer (LAK), cytolytic T lymphocytes (CTL) (against specific and nonspecific targets), and macrophage tumoricidal and tumoristatic activities were measured. rM IFN-gamma and pICLC had therapeutic activity and immunomodulatory activity in most assays of immune function examined. Specific CTL activity of pulmonary parenchymal mononuclear cells (PPMC), but not in splenocytes or peripheral blood lymphocytes (PBL), during week 3 and not during week 1, correlated with the therapeutic activity of rMu IFN-gamma and of pICLC. Macrophage tumoricidal activity in PPMC, but not in alveolar macrophages, also correlated with the therapeutic activity of rMu IFN-gamma, but the opposite was true for the therapeutic activity of pICLC. NK activity of PPMC, but not of splenocytes or PBL, during week 1 correlated with the therapeutic activity of pICLC; in contrast, NK activity at any site did not correlate with the therapeutic activity of rMu IFN-gamma. LAK activity at any site did not correlate with the therapeutic activity of either agent.  相似文献

Proteinase K, the extracellular serine endopeptidase (E.C. 3.4.21.14) from the fungus Tritirachium album limber, is homologous to the bacterial subtilisin proteases. The binding geometry of the synthetic inhibitor carbobenzoxy-Ala-Phechloromethyl Ketone to the active site of proteinase K was the first determined from a Fourier synthesis based on synchrotron X-ray diffraction data between 1.8 Å and 5.0 Å resolution. The protein inhibitor complexes was refined by restrained least-squares minimization with the data between 10.0 and 1.8 Å. The final R factor was 19.1% and the model contained 2,018 protein atoms, 28 inhibitors atoms, 125 water molecules, and two Ca2+ ions. The peptides portion of the inhibitor is bound to the active center of proteinase K by means of a three-stranded antiparallel pleated sheet, with the side chain of the phenylalanine located in the P1 site. Model building studies, with lysine replacing phenylalanine in the inhibitor, explain the relatively unspecific catalytic activity of the enzyme.  相似文献

对大肠杆菌生产的不同型别的基因工程干扰素rIFN-α1(α1)、rIFN-αA(αA)、rIFN-β17ser(β17ser)和rIFN-γ(γ),以及自然人白细胞干扰素nIFN-αco,在不同细胞上对不同病毒的抗病毒活性做了比较研究。证明:①α1抗病毒作用的细胞谱较广,尤其在牛肾MDBK细胞和猪肾PK细胞上有很高的活性,分别为在人细胞上的29倍和7倍。β17ser和γ在异种细胞上活性极低,在鼠、猪和牛肾细胞上的活性为人细胞上的1~2%以下。②5种干扰素对麻疹、CoxB1、Sindbis、腺病毒7型和Ⅰ、Ⅱ型单纯疱疹病毒的抗病毒活性无明显差异。但不同病毒对干扰素的敏感性有明显差别,以Sindbis病毒为最敏感,7型腺病毒最不敏感。③5种干扰素对流行性出血热病毒均有明显的抗病毒作用,尤以人α1型和β干扰素作用最强,α1对出血热病毒的抗病毒活性是对滤泡性口膜炎病毒(VSV)的1/2.85,人β干扰素是对VSV的1/4.1。上述结果为人基因工程干扰素的临床应用提供了实验依据。  相似文献

Summary Four experimental formulations ofBacillus sphaericus Neide (2362 isolate) were evaluated for larvicidal activity against culicine and anopheline larvae in several natural and artificial habitats. A granular formulation (5% primary powder) was tested against natural populations of mosquitoes in two simulated habitats in Florida and in maturing and reflooded rice fields in Louisiana. Larvae ofCulex quinquefasciatus Say were reduced by 97 and 99% after application of the granules at the rate of 10 kg/ha to polluted tanks and 2.5 kg/ha to sod-lined potholes, respectively. Anopheline andPsorophora columbiae (Dyar and Knab) larvae were reduced by 68 and 92–100%, respectively, after application of 5 kg granules/ha to rice fields. A flowable concentrate (12.8% primary powder) applied to unpolluted and organically enriched habitats in Florida at 0.25 kg/ha reduced populations ofCulex spp. by 93–100% and 99%, respectively. Sustained-release briquets (5% primary powder) applied at the rate of one half briquet/1.8 m2 sod-lined potholes reduced larval populations ofCx. quinquefasciatus by 88–95% for up to 2 weeks in open sunlight. Sustained-release pellets (30% primary powder) applied to small woodland pools in Memphis, TN at the rate of four pellets/pool virtually eliminated larval populations ofCx. restuans Theobald for over 8 days. Variable persistance of larvicidal activity was noted for the other treatments depending on the formulation, target species and habitat.  相似文献

This substrate was prepared by incorporating black drawing ink into casein and heating this complex at 200°C for 4 h. In connection with our discovery of a strong immunosuppressive activity of cyclolinopeptide A (CLA), we investigated immunosuppressive properties of antamanide and a number of its analogues, including symmetrical antamanide, and compared them with the activities of cyclosporin A and CLA. The peptides were investigated by using plaque forming cell (PFC), graft-versus-host (GvH), delayed type hypersensitivity (DTH), and autologous rosette formation cell (ARFC) tests. Antamanide and symmetrical antamanide exhibit an immunosuppressive activity lower than CLA. Linear antamanide fragments are also active. At higher concentrations of the latter peptides, toxic effects occur.  相似文献

The entry into, and exit from, mitosis are controlled by a universal M-phase promoting factor (MPF) composed of at least p34cdc2 and a cyclin. Embryonic systems are convenient for studying the association and dissociation of the active MPF complex because oocytes and eggs are naturally arrested at a specific point of the cell cycle until progression to the next point is triggered by a hormonal signal or sperm. In amphibians, eggs prior to fertilization are arrested at metaphase 2 of meiosis due to the presence of a stabilized MPF complex. Fertilization (egg activation) produces a transient increase in intracellular free Ca2+, a propagating Ca2+ wave, that specifically triggers the destruction of cyclin, leading to MPF inactivation and entry into the first embryonic inter-phase. We have recently shown that intracellular pH (pHi) variations in amphibian eggs, a large increase at fertilization and small oscillations during the embryonic cell cycle, were temporally and functionally related to the corresponding changes in MPF activity. In addition, the recent finding that the pHi increase at fertilization in Xenopus eggs is a propagating, Ca(2+)-dependent pH wave which closely follows the Ca2+ wave, together with the absence in the egg plasma membrane of pHi-regulating systems responsible for that pHi increase, suggest the existence of cortical or subcortical vesicles acidifying in the wake of the Ca2+ wave, thus producing the pH wave.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
